Enzyme Digestion (time for lysis: 15-30 min per 1g cell)
1) 理論(이론)
lysozyme을 이용, cell lysis
TE buffer: 50mm Tris-HCl (8.0), 10mM EDTA
2) 사용법
1. 1g의 cell 당 3ml의 TE buffer washing, 20-37℃ 배양.
2. cell suspension 5ml 당 1 mg의 lysozyme (10 mg/ml stock)을 첨가.
3. 20-37℃ 10-20 min shaking

순서
다. Whole cell protein을 얻을 때 적당하다. RIPA는 Radioimmunoprecipitation assay buffer의 약자이다.
2) 조성
- pH 8.0
- Tris 50mM
- NaCl 150mM
- SDS 0.1% (m/v)
- Na-Deoxychlolate 0.5% (m/v)
- Triton X-100 or NP-40 1% (v/v)
- 사용하기 직전에 protease inhibitors 추가
Sonication (time for lysis: 2 min per 1g cell)
1) 理論(이론)
Sonicator로 cell wall에 진동을 발생시켜 물리적으로 파괴.
2) 사용법
Cell을 선택한 buffer로 wasing 후 sonication
위상차 현미경 하에서 sonication 강도와 시간에 따른 cell의 파괴 정도를 check하여 최적 조건을 찾아둔다.
